Install a Safari Reader hack
If you’ve downloaded a Reader hack, such as Antique, this is how you install it and put it to use.
Update for Lion:
The instructions below are still relevant, but the location of Reader.html has changed. Instead of looking in the Safari application bundle, you’ll need to look for it at /System/Library/PrivateFrameworks/Safari.framework/Versions/A/Resources/Reader.html. You’ll need Administrator (or sudo) privileges to modify it. Thanks to Jeremy Lu for the pointer.
Preparing Reader.html
First, quit Safari if it’s running.
The hack you downloaded should include a file titled “Reader.html”. Put this file in a safe location, such as your “Documents” folder. When Safari is upgraded next time a new version is released, your hack will be overwritten and you’ll need this file again.
Open the Safari Application bundle

Locate Safari.app in your Applications folder. It may appear as just “Safari” if you don’t have your Finder set to show extensions. Right click on the application and choose “Show Package Contents.”
Locate Reader.html and back it up

Inside of the application bundle, locate Contents > Resources > Reader.html. Move this file to a safe place as a backup, but make sure it’s removed from the folder. Enter your System Password if asked.
Copy the new Reader.html into the Safari application bundle
Copy the new Reader.html into the Resources folder in the Safari application bundle, where the original was before you moved it.
You can now start Safari and test the new Reader hack by going to a page which triggers Reader and loading it!
